    针对微囊藻在自然环境中常聚集成团、难以统计分析的问题,探索了不同功率和处理时间下超声波对微囊藻群体打散状况,并通过浮游植物流式细胞仪和EasyClus分类计数软件对微囊藻数目进行精确统计。结果表明:随着超声波功率和处理时间的增加,微囊藻打散效果呈现先上升后下降的趋势。在480W超声波功率处理96s后,微囊藻颗粒数和红色总荧光值达到最高,FWS前向散射值最小。480W超声波打散处理96s后发现微囊藻细胞破碎,且打散时间继续增加,细胞破碎愈发严重。因此确定,微囊藻群体的最佳超声波打散功率为480W,最佳处理时间为72s。综合镜检和EasyClus的分析结果证明,本方法准确且高效,可用于实际藻类的快速监测。

    Abstract:

    Microcystis present as colonies in natural environment, which make it difficult to calculate Microcystis cells number. This paperexplored the disruption of Microcystis in different ultrasonic power and treatment time, and Microcystis cells number was quantified by CytoSenseand EasyClus. The experimental results show that with the increase of ultrasonic power and treatment time, the effect of dispersion improved firstand then decreased. The number of Microcystis particles and the total value of red fluorescence reached the highest after 96 s ultrasonic treatment(480W), at the same time, FWS forward scattering value reached minimum, and Microcystis cells were disrupted. Also under 480W, with theincrease of treatment time, more Microcystis cells were disruped. Therefore, it was determined that the optimal ultrasonic dispersion power forMicrocystis colonies was 480W, and the optimum treatment time was 72s. The results of microscopy counting and EasyClus analysis show that thismethod is accurate and and efficient, which can be used for rapid algae monitoring.
